Applications of Super Resolution Expansion Microscopy in Yeast

چکیده

Super-resolution microscopy includes multiple techniques in optical that enable sub-diffraction resolution fluorescence imaging of cellular structures. Expansion (EXM) is a method physical expansion to obtain super-resolution images biological sample on conventional microscopy. We present yeast organelles, applying the combination and ExM techniques. When preparing pre-expanded samples, methods lead breakage dividing cells difficulties studying division-related proteins. Here, we describe an improved preparation technique avoids such damage. with Airyscan structured illumination (SIM) collected sub-cellular structural nuclear pore complex, septin, a-tubulin yeast. Our well-suited for study

Ex-STORM: Expansion Single Molecule Super-resolution Microscopy

We present critical improvements to recently invented Expansion Microscopy (ExM), which resolve the incompatibility between ExM and single molecule super-resolution microscopy STORM. Specifically, the improved ExM circumvents the massive, 50-100%, bleaching of fluorophores during sample preparations, and preserve the efficiency of enzymatic oxygen scavenging systems in expanded samples. These i...

Super-resolution Microscopy – Applications in Plant Cell Research

Most of the present knowledge about cell organization and function is based on molecular and genetic methods as well as cytological investigations. While electron microscopy allows identifying cell substructures until a resolution of ∼1 nm, the resolution of fluorescence microscopy is restricted to ∼200 nm due to the diffraction limit of light. However, the advantage of this technique is the po...

Super-Resolution Localization Microscopy

Light microscopy techniques have been vital to our understanding of biological structures and systems since their invention in the late 16 Century. However, conventional light microscopy techniques are limited by the diffraction limit of light, preventing us from seeing anything smaller than approximately 200 nm in x and y and approximately 500 nm in z. Many molecules and structures of interest...

Super-resolution microscopy of mitochondria.

Mitochondria, the powerhouses of the cell, are essential organelles in eukaryotic cells. With their complex inner architecture featuring a smooth outer and a highly convoluted inner membrane, they are challenging objects for microscopy. The diameter of mitochondria is generally close to the resolution limit of conventional light microscopy, rendering diffraction-unlimited super-resolution light...

Multi-plane super-resolution microscopy

Understanding cell functions is the major goal of molecular biology, which intends to elucidate the interactions between biomolecules at a subcellular level. One of the widely used techniques in molecular biology is fluorescence microscopy, which offers high specificity and sensitivity at the submicrometer spatial scale but is limited by diffraction to about 200nm lateral resolution, which is i...
